When freezing cells, tissues and biological samples, it is not only the final temperature that matters, but above all the way the sample cools down. A thermocouple sensor measures the actual sample temperature inside a straw or cryovial, while a chart recorder captures the freezing curve on paper. This gives you verifiable proof that the freezing protocol was carried out correctly. During controlled-rate freezing in a CryoMed controlled-rate freezer, the chamber temperature follows a programmed profile, but the temperature inside the sample itself always lags behind it. In particular, the moment the heat of crystallisation is released (the phase change from liquid to solid) causes a brief temperature spike inside the sample. To maximise cell survival, that spike must be kept under control.
That is why you use a thermocouple sensor to measure the temperature directly inside a representative test sample, a straw or cryovial identical to the rest of the batch. The freezer can then adjust its cooling based on the actual sample temperature and not just on the chamber temperature. The recorder captures both curves, so you can afterwards demonstrate that the protocol ran as planned.
