High capacity for large sample series

Firstly, the SPD210 processes up to 200 × 1.5-2.0 mL microtubes per run with the RH200-12 rotor. In addition, the system accepts 192 × 1-dram glass vials, 48 × 50 mL conical tubes, 88 × 16 × 125 mm tubes, 50 × 20 mL scintillation vials and 8 × 100 mL pear-shaped flasks. Moreover, microplate rotors are available for shallow well and deep well plates. As a result, the SPD210 is suitable for laboratories processing large series in a compact footprint.

Modular design for maximum flexibility

Firstly, the SPD210 is deliberately designed as a standalone concentrator unit. As a result, you choose which cold trap and which vacuum pump best matches your lab. Moreover, Thermo recommends two configurations. The first option is the Semi-Integrated System with UVS450 Universal Vacuum Source and ANT100 post-trap. The second option is the Component High Vacuum System with separate RVT450 cold trap, VLP120 vacuum pump, SCT120 chemical trap and DVG50 vacuum gauge. Consequently, you assemble a system that exactly matches your existing infrastructure.

Designed for non-aggressive organic solvents

Firstly, the SPD210 is designed for non-aggressive organic solvents. Typical solvents include methanol, acetonitrile, ethanol, isopropanol, n-butanol, isobutanol, n-propanol, n-hexane, n-heptane, pyridine, ammonium hydroxide and water. In addition, the system works with combinations of water+ACN+MeOH with low concentrations of acid (0.1% TFA). Therefore, the instrument is ideal for HPLC fractions and reversed-phase SPE eluent applications. However, for aggressive solvents such as DMSO, methylene chloride or strong acids and bases, you choose the SPD300DDA high capacity DDA variant.

3 modifiable programs plus two separate timers

Firstly, the control panel offers three modifiable factory programs. By default, these are set to Water (45°C, 2 hour heat time, 2 hour run time), Low Boiling ACN/MEOH (65°C, 2 hour heat time, 2 hour run time) and HPLC (no heating, 2 hour run time). In addition, you set the chamber temperature per program between 35°C and 65°C in 5°C increments. Alternatively, you select “no” for no heating at all. Moreover, the SPD210 works with two independent timers (heat time and run time). Each timer can be set from 1 minute to 9 hours 59 minutes, or to continuous (CCC) for the heat time.

USB data export for traceability

Firstly, the SPD210 features a USB type A/B port that exports live run data to an external computer. Subsequently, the system sends the live chamber temperature, remaining heat time and remaining or elapsed run time every minute. Moreover, the data arrives in CSV format at a fixed baud rate of 115200. As a result, you easily meet GMP and GLP reporting requirements for pharmaceutical and biotech applications.

Safety and ease of use

Firstly, the glass cover with safety interlock locks automatically during a run. Consequently, the cover opens only when the rotor has fully stopped and the vacuum has been released. In addition, the SPD210 features a hydraulic damper that lets the cover open and close smoothly. Finally, a Cover Lock Emergency Release Tool is supplied as standard for removing samples during a power failure.

Two recommended system configurations

Firstly, Thermo Scientific offers two complete kit options for the SPD210. The first is the SPD210P1-230 kit with RVT5105 ultra-low temperature vapor trap (-104°C), vacuum pump, RH200-12 rotor (200 × 1.5 mL), digital vacuum gauge, universal tubing, chemical trap, disposable cartridge, glass condensation flask and CryoCool heat transfer fluid. Secondly, the SPD210P2-230 kit with UVS450 Universal Vacuum Source (-55°C), tubing kit, CryoCool and RH200-12 rotor.

Tips for faster drying

Firstly, Thermo offers several practical tips for the highest drying speed. To begin, you use glass tubes and vials instead of plastic, because glass conducts heat better. In addition, you preheat the SpeedVac chamber and rotor before loading samples. Moreover, rotors with metal tube carriers (RH12-20, RH8-18) help with faster heat transfer. Finally, you prewash plastic tubes with methanol to remove materials that inhibit evaporation of aqueous samples.

What’s in the box?

Firstly, the SPD210 is available in three configurations. The first is the standalone concentrator unit with power cord and Cover Lock Emergency Release Tool. Secondly, the complete SPD210P1-230 kit for HPLC and SPE applications with RVT5105 vapor trap, vacuum pump, rotor, vacuum gauge and accessories. Thirdly, the SPD210P2-230 kit with UVS450 Universal Vacuum Source and RH200-12 rotor. In addition, for optimal vacuum pump maintenance with aggressive solvents, Thermo recommends a VPOF110 oil filter.

Which laboratories use the SPD210?

Firstly, the SPD210 is used in life sciences, biotech, organic chemistry, drug discovery, peptide synthesis and the pharmaceutical industry. Typical applications include HPLC fractions with water and 0.1% TFA, reversed-phase SPE eluent processing, protein and peptide concentration in large series, DNA/RNA precipitation and metabolite research. What is the SpeedVac SPD210 used for?

The SPD210 is used for high-throughput concentration and drying of samples in non-aggressive organic solvents. Typical applications include HPLC fractions with water and 0.1% TFA, reversed-phase SPE eluent processing, protein and peptide concentration, DNA/RNA precipitation and metabolite research in large series.

What is the difference between the SPD210 and the SPD2030?

Both are high capacity SpeedVac models with the same capacity of 200 microtubes per run. However, the main difference is the modular design. The SPD2030 is a fully integrated system with built-in cold trap, pump and vacuum gauge. By contrast, the SPD210 is modular, which means you combine the concentrator unit with a separate cold trap and vacuum pump. As a result, the SPD210 offers more flexibility for applications requiring specific cold trap temperatures, such as the -104°C RVT5105 for aggressive solvents.

What is the difference between the SPD210 and the SPD300DDA?

Both are high capacity modular SpeedVacs of comparable size. However, the main difference is chemical resistance. The SPD210 is intended for non-aggressive organic solvents such as methanol, acetonitrile, ethanol and water. By contrast, the SPD300DDA is industrial strength and resistant to aggressive solvents such as DMSO, methylene chloride, TFA and strong acids and bases. Therefore, for combinatorial chemistry or aggressive applications you choose the SPD300DDA.

For example, water, acetonitrile, methanol, ethanol, isopropanol, n-butanol, isobutanol, n-propanol, n-hexane, n-heptane, pyridine and ammonium hydroxide. In addition, the system works with combinations of water+ACN+MeOH with low concentrations of acid (0.1% TFA). However, for aggressive solvents such as DMSO, methylene chloride, strong acids or bases, you choose the SPD300DDA.

What is the difference between SPD210, SPD210P1 and SPD210P2?

All three contain the same SPD210 concentrator unit. The SPD210-230 is the standalone unit with Cover Lock Emergency Release Tool. The SPD210P1-230 is a complete kit for HPLC and SPE applications with RVT5105 ultra-low temperature vapor trap (-104°C), vacuum pump, RH200-12 rotor, digital vacuum gauge, universal tubing, chemical trap, disposable cartridge, glass condensation flask and CryoCool. The SPD210P2-230 is a kit with UVS450 Universal Vacuum Source (-55°C), tubing kit, CryoCool and RH200-12 rotor.

Which cold trap do I need with the SPD210?

You choose between two recommended options. Firstly, the RVT5105 ultra-low temperature vapor trap (-104°C), suitable for lyophilization and low-boiling solvents. This is included as standard in the SPD210P1 kit. Secondly, the UVS450 Universal Vacuum Source with -55°C cold trap, which is included in the SPD210P2 kit. Alternatively, you choose a component setup with the RVT450 (-50°C) plus a separate VLP120 vacuum pump.

How many samples can the SPD210 process per run?

Depending on the rotor, the SPD210 processes up to 200 × 1.5-2.0 mL microtubes (RH200-12), 192 × 1-dram glass vials (RH192-15), 48 × 50 mL conical tubes (RH48-50U), 88 × 16 × 125 mm tubes (RH88-16-125), 50 × 20 mL scintillation vials (RH50-28-60) or 8 × 100 mL pear-shaped flasks (RH8-200). Furthermore, microplate rotors are available for shallow well (12 plates) and deep well (4 plates) applications.

Is data export possible?

Yes, via the USB type A/B port you export live run data at a fixed baud rate of 115200. Every minute, you receive the live chamber temperature, remaining heat time and run time in CSV format. As a result, you easily meet GMP and GLP reporting requirements for pharmaceutical and biotech applications.

Which programs come standard on the SPD210?

The SPD210 is supplied with three modifiable factory programs. Firstly, Water (45°C, 2 hour heat time, 2 hour run time). Secondly, Low Boiling ACN/MEOH (65°C, 2 hour heat time, 2 hour run time). Thirdly, HPLC (no heating, 2 hour run time). In addition, you customize and save each program by holding the Program button for 3 seconds.

How can I increase the drying speed?

Thermo provides several tips for faster drying. Firstly, you use glass tubes and vials instead of plastic. Secondly, you preheat the SpeedVac chamber and rotor before loading samples. In addition, rotors with metal tube carriers (such as RH12-20 or RH8-18) help with faster heat transfer. Finally, you prewash plastic tubes with methanol to remove materials that inhibit evaporation of aqueous samples.

Which power supply does the SPD210-230 require?

230 VAC / 50 Hz, 6 amperes. The plug is a SCHUKO standard European plug and fits directly into Dutch wall sockets. The fuse is a 6 A, 250 VAC time-lag fuse. Note that the SPD210 draws more current than the medium capacity SpeedVacs (3A for SPD120/SPD140DDA), so make sure you have a suitable outlet.

What is the temperature range of the SPD210?

The SPD210 operates from 35°C to 65°C in 5°C increments, or “no” for no heating. This is slightly lower than the medium capacity SpeedVacs which go up to 80°C. The lower maximum temperature is optimized for protecting biological samples during long runs with large sample volumes.
